Ribosomal Synthesis of Macrocyclic Peptides for Chronic Myelogenous Leukaemia Treatment
نویسندگان
چکیده
Introduction In Nature, the vast majority of polypeptides are synthesized through translation of the messenger RNA (mRNA) by the joint action of the ribosome and the transfer RNA (tRNA). The mRNA is a transcribed copy of the gene that carries genetic information in the form of series of three-nucleotide sequences referred as ‘codon’. Each codon corresponds to a specific amino acid or a termination signal. Ribosomal translation of the mRNA relies on the specific hybridization between the complementary bases of the codons of the mRNA and the anticodons of the tRNA which is specifically conjugated to its cognate amino acid by an enzyme called aminoacyl tRNA synthase (aaRS). Bioinspired Strategy for the Ribosomal Synthesis of Thioether-Bridged Macrocyclic Peptides in Bacteria
Inspired by the biosynthetic logic of lanthipeptide natural products, a new methodology was developed to direct the ribosomal synthesis of macrocyclic peptides constrained by an intramolecular thioether bond. As a first step, a robust and versatile strategy was implemented to enable the cyclization of ribosomally derived peptide sequences via a chemoselective reaction between a genetically enco...

متن کاملPregnancy Outcome of Two Patients with Chronic Myelogenous Leukemia Treated with Imatinib
Although chronic myelogenous leukemia in pregnancy is rare, its management and treatment is more difficult and complicated.Treatment of patients with chronic myelogenous leukemia includes bone marrow transplantation, however in less than 30% of patients the donor’s organ would be accepted. To this end, cytotoxic therapy is considered as an alternative therapeutic option. This option provides sa...
متن کاملCoexpression Pattern Analysis of NPM1-Associated Genes in Chronic Myelogenous Leukemia
BACKGROUND Nucleophosmin 1 (NPM1) plays an important role in ribosomal synthesis and malignancies, but NPM1 mutations occur rarely in the blast-crisis and chronic-phase chronic myelogenous leukemia (CML) patients. The NPM1-associated gene set (GCM_NPM1), in total 116 genes including NPM1, was chosen as the candidate gene set for the coexpression analysis. We wonder if NPM1-associated genes can ...
متن کاملChronic myelogenous leukemia in ulcerative colitis
Patients diagnosed with inflammatory bowel disease are known to be at an increased risk of colorectal and liver cancers and of leukemia. In fact, several forms of primary and secondary hematological malignancies were rarely observed during the clinical course of inflammatory bowel diseases. The causal relationship, if any, remains undetermined. Patients needing a prolonged treatment with immu n...
